摘  要:本文应用透射电子显微镜观察了在林业方面具有开发利用价值的苏云金芽孢杆菌SFZZ03菌株在牛肉膏蛋白胨液体培养基中的发育历期。本实验在发酵的不同时间进行取样,经过电镜样品前期处理,在透射电镜下观察SFZZ03菌株的细胞分裂,芽孢、伴孢晶体的形成,不仅详细阐述了SFZZ03菌株的发育过程,而且初步判断了伴孢晶体、芽孢的最大产量期,为今后的大规模生产确定种子罐培养时间、发酵罐培养时间提供了参考数值。同时也为筛选高产量伴孢晶体菌株提供了一种有效的测定方法。

Observation on development process of Bacillus thuringiensisSFZZ-03

Abstract  This research focuses on the development duration of Bacillus thuringiensisSFZZ-03 which has Exploitation and utilization value in forestry in LB medium. In this experiment, samples were prepared at different times of fermentation, and then the development duration of Bacillus thuringiensisSFZZ-03 which contains cell division, sporulation and parasporal crystal formation were observed. This research not only provides details on development process of Bacillus thuringiensisSFZZ-03, but preliminarily analyzes the mass production of parasporal crystals and spores. It provides a reference value in determining tank culture time and fermenter time for large scale production in the furthure. It also provides an effective measuring method for screening high yield strains with parasporal crystals.
